On his first appeareance in Showcase #4 he fought two villains: Turtle Man and ̶G̶r̶a̶n̶t̶ ̶M̶o̶r̶r̶i̶s̶o̶n̶ Mazdan
The Shade was the first Jay Garrick supervillain; the first Barry Allen supervillain was Turtle Man who was based on the Garrick villain The Turtle.
The list in is missing a bunch of people before Grodd and The Pied Piper. Captain Cold and Mirror Master were both earlier plus Mazdan, who appeared in the same issue as Turtle Man and is technically the first original supervillain original to Barry.

That was October 1956, right in Barry's origin issue and three years before Grodd. The Turtle being Barry's first enemy is even a plot point in the Flash: Year One story of the current run.

Killer Moth was Batgirl's first villain. Being defeated by a woman in her first appearance is undoubtedly why writers started treating him like a joke.
