Tifa is also strong enough to pick the sword up no problem, but she was trained by the world's strongest man for years, so that makes sense.

More magic. Its literally all magic. Mako is just concentrated earth/soul magic. When it dries and solidifies it turns into materia which are magic stones.

You might be misremembering. I think it's actually Sephiroth's sword that she picks up. Which is still pretty unwieldy, but it definitely wouldn't weigh 50+ kg like the Buster Sword would. I also think that might be before she was taken by Zangan for her secret training.

As far as Cloud goes, yeah I guess you're right about that one. Although it's worth mentioning that he doesn't do much with it other than drive it into the back of an unsuspecting man. A really strong normal guy could probably lift the Buster Sword, he just wouldn't be able to fight with it.
