Nah, your eyes are relaxed like your'e looking in the distance even though you have lenses right up in your face.
The only games I've had any strain with were redout, which is the fastest, most colorful arcade racer I know, or borderlands 2 VR, which has tons of colors and explosions all the time. Even then it's minimal, and feel relief just taking a short break or closing eyes for a minute every couple hours or something like that.
