Or will they use the skill/knowledge they demonstrated in their series, and will win/lose based on the skill difference between that, and their opponent?

I imagine the Sakamoto answers believe plot armor is the ultimate strength and put their faith in that, both in this game and in their reading and watching. Key sentence here. Since Sakamoto doesn't really have a particular niche but is just talented at everything, he'd make the safest bet. Although he could still be defeated depending on what the game actually entails, even then he's probably the guy on the list most likely to put your own safety before his own. He might not win, but would probably pull off some dumb loophole to get you out of trouble.

This is something literally no other human on the planet will ever be able to do. Even the world chess champion gets annihilated playing them.

I don't know all these other characters, but if it's a chess-like game, I want Shiro. If it's a game that allows cheating, I probably want her as well, because they do that too.
If it's a random bullshit asspull game that's literally impossible to predict/play? I still want her, because they did that too.

So my point is that if we accept the premise and the canon, then Shiro will win at everything, by the premise.
