That would be pretty difficult to adapt because it can't be divided by chapters like one hundred years of solitude, it must be a movie and at least 3 hours of running time.
If you insist on adapting juan rulfo then I would love an adaption of el llano en llamas like the junji ito anime.

How the heck would adapting 100 years of solitude even work? How do you even adapt the atmosphere to an animated medium? You need to have and knowledge of colombian history, society and culture to really understand what happens in the book and what it means, would any Japanese studio ever give a shit?
An anime adaptation would be basically Jojo lite with less muscle guys posing and more melancholic old men making fish sculptures

The first time I read it I didn't know about any of the stuff that you mentioned and still don't know a lot about colombian society or culture outside of Pablo Escobar, and still enjoyed it.
You are right to have your worries about how are they going to adapt a book with this complexity, but all adaptions are not 100% true to the original work and when you do this transition from a written to a visual medium then the changes are going to be radical.
I have seen several anime where magical realism worked fine like Jinrui wa Suitai Shimashita so I have faith that if there is someone that can do a good adaption of the book into a movie or series, it's the japanese.

How many anime are actually adaptations of proper novels and not light novels/web novels?
The first that comes to mind for me is LoGH.
Preferably directed by Miyazaki but he probably despises Mishima

Also many of the classics are, like World Masterpiece Theater and the competitors

Oh right, I forgot that Shinsekai Yuri was adapted from a proper novel as well, but it makes sense considering how much better the plot and worldbuilding is compared to the average anime.

Anything by Kobo Abe would work really well as an anime. I want to see more Kafka and Lovecraft done too. Dunwich Horror had an EXCELLENT puppet movie.
