It's obvious that he has a serial number under his eye considering he's already employed as a blade runner. The mystery comes from the clues that seemingly contradict the fact that he is a replicant.

Why wouldn't they engineer replicants to have blue skin or barcode tattoos on their face or something, to make them stand out if they get back to Earth? Make them obvious.

The protagonist and the audience not knowing if he's human or a replicant encourages both to examine what conciousness means. The film fails if he is a replicant, because then his recognition of Rachel and Roy as having humanity is rendered meaningless. It only has meaning if a human, conscious of being human, recognizes humanity in two replicants, both of whom the human is conscious of as being replicants. What's the point of Roy asserting his humanity, through mercy and a display of sincere emotions, to Rick if Rick is also a replicant?
Furthermore, through Rachel and Roy, the audience can still be prompted to consider strong AI without the need for Rick to be non-human.
