Tolkien himself said it was unclear. The end-of-book preview in TFOTR says that they are Orthanc and Minas Morgul, but it’s not known if Tolkien wrote those himself. Anyway one of them is almost certainly Orthanc.

Tolkien never cared much for the titles his editor suggested after splitting the book into three but he did say that he considered the titular towers to be Orthanc and Barad-dur.

I do think that the title works better as Isengard and Mordor both fucking Middle Earth.

No, those are Minas Morgul and Orthanc. It's clearly seen in Tolkien's illustration for the cover of TTT - on the left there's a white tower with a crescent (Minas Ithil (before it was captured and called Minas Morgul) means Tower of the Rising Moon) and on the right there's a black tower with a circling stair with prongs on top - which is clearly Orthanc. Plus, you can see the white hand (Saruman's symbol) underneath and a star (it doesn't have any concrete meaning but it probably refers to Saruman's habit of observing stars on Orthanc's top). And at last where's the winged Nazgûl flying to interrogate Saruman/retrieve his spoils. I'm not sure about significance of the nine circles at Minas Morgul - probably it's the nine Nazgûl.

Cirith Ungol is a valley in the Ephel Duath(Mordor mountain range) at the end of it is the tower of Cirith Ungol. Minas Morgul is at the other end. Kind of.
