Some of these I get from context clues like the Damien Hirst, but I have to google them to get the real reference. I've always glossed over the painter names. Shame as a Greek I didn't really register El Greco. Spanakopita on the other hand was a treat of an episode.

In Escape to House of Mummies pt 2 when they're testing the shrink ray on Billy and White shrinks his head he says "Nice shot, William Burroughs" Burroughs was a beat generation writer and artist who was convicted of manslaughter in absentia for accidentally shooting his wife in the head while drunkenly horsing around with a gun. >Damien Hirst There's a lot of visual jokes, too. The dissected shark that Wide Whale steals for Dugong in their origin flashback is based on a Hirst piece. Also Scare Bear was based on a performance art piece.
I hadn't heard of the slang "battle ax" when I originally watched season 6 so that was a fun little realization. I also only somehow got the double joke with Headshot's name like a week ago.

I think it's a mix of reasons. Characters have funny voices, music tends to play over what they say, and the writing itself doesn't make it easy to figure out what they're saying from context.
Pic-related is the first thing that came up when I searched for "venture bros quotes". You see characters speaking in funny accents and using terms like "birth crevasse" you'd never hear elsewhere. Even if you know what a crevasse is you might incorrectly group up "birthcrevasse" as one word and not know what the fuck she's talking about. >"What the fuck does the word 'birthcrevasse' mean? Oh 'BIRTH crevasse' okay..."
My parents aren't native English speakers so as a young child I guess I was sort of taught a mix of English and Spanish. Either way English has by far been my primary language most of my life (wouldn't even consider myself fluent in Spanish at this point).
