We don't really know if Netflix Isaac is gay but the game one definitely was and can't imagine Netflix changing that. He did say he loved his former master but could've been in a familial way

Once Hector’s hit rock bottom the only place to go is up, just gotta get Lenore to fall in love with him/become his world like Dracula was for Isaac before Isaac shows up for the slaughter. Then you can have a parallel character development arc but with the new version of Isaac’s sister giving him something positive to live for past the revenge.
